A recombinant HEK293 cell line constructed to express human PDE10A2 (phosphodiesterase 10A, accession number NM_006661). This cell line features N-terminal His-tagged human PDE10A2, with stable expression confirmed by Western blotting. The function of PDE10A2 was confirmed using a cAMP detection assay. Activation with forskolin led to increased cAMP levels in parental HEK293 cells, while cells overexpressing PDE10A showed reduced cAMP accumulation. Inhibition of PDE10A activity restored cAMP levels.
Drug screening and biological assays
PDE10A (Phosphodiesterase 10A) can hydrolyze both cAMP and cGMP. It is highly expressed in the striatum of the brain and is involved in the regulation of neuronal activity and motor control, as well as in other physiological and pathological processes.
