Introduction

The Human CD16a (158V) (Luc) Jurkat Reporter Cell has been created to express both the NFAT response element driving luciferase expression and the full-length human CD16a receptor (Uniprot: P08637) with a Valine (V) mutation at amino acid 158. This receptor exhibits a higher affinity for IgG1 and IgG3 isotypes compared to CD16a-158F. The cell line can be used to evaluate the ADCC activity of antibodies in the presence of corresponding target cells.

Background

The FCGR3A gene encodes a low-affinity receptor for the Fc region of immunoglobulin G (IgG). It is expressed on the surface of natural killer (NK) cells, macrophages, and some T cells. This receptor plays a crucial role in antibody-dependent cellular cytotoxicity (ADCC), where it binds to the Fc portion of antibodies attached to target cells, leading to the activation of immune cells and the destruction of the target cells. It is also involved in the regulation of immune responses and inflammation.
