Introduction

The Human 4-1BB (Luc) HEK293 Reporter Cell was engineered to express the NF-KB signaling response element and the full-length human 4-1BB receptor (Uniprot: Q07011). This cell line can drive luciferase expression in response to 4-1BB ligand or agonist antibody stimulation. In the absence of agonist antibody or 4-1BB ligand, the 4-1BB receptor is inactive, resulting in low luminescence. In the presence of agonist antibody or 4-1BB ligand, dose-dependent 4-1BB pathway-activated luminescence can be detected. This reporter cell can also be used to test whether agonist antibodies enhance activity in an Fc纬R-dependent manner.

Application

Screen for ligands or agonist antibodies that can bind and activate 4-1BB

Background

4-1BB (also known as CD137) is a costimulatory molecule that enhances T cell proliferation and survival upon engagement with its ligand, 4-1BBL. It plays a crucial role in immune responses against tumors and infections.
