The HEK293-Human 4-1BB (Luc)-Reporter Cell is engineered to express both the NF-KB signaling response element and the receptor full-length human 4-1BB (Uniprot: Q07011), which can drive luciferase expression systems upon 4-1BB ligand/agonist antibody stimulation. In the absence of agonist antibody or 4-1BB ligand, the 4-1BB receptor is not activated, and luminescence signal is low.

4-1BB (TNFRSF9, CD137) is a co-stimulatory receptor expressed on activated T cells, NK cells, and dendritic cells. It binds to 4-1BB Ligand (TNFSF9) on APCs, promoting T cell proliferation, survival, and cytokine production. 4-1BB agonists are being tested in cancer immunotherapy.
