The Human CD40 (Luc) HEK293 Reporter Cell was engineered to express the NF-KB signaling response element and the full-length human CD40 receptor (Uniprot: P25942-1). This cell line can drive luciferase expression in response to CD40 ligand or agonist antibody stimulation. In the absence of agonist antibody or CD40 ligand, the CD40 receptor is inactive, resulting in low luminescence. In the presence of agonist antibody or CD40 ligand, dose-dependent CD40 pathway-activated luminescence can be detected. This reporter cell can also be used to test whether agonist antibodies enhance activity in an Fc纬R-dependent manner.

CD40 is a costimulatory molecule on antigen-presenting cells that interacts with CD40L on T cells to enhance B cell activation and antibody production.
