The HEK293-Human CD40 (Luc)-Reporter Cell is engineered to express both the NF-KB signaling response element and the receptor full-length human CD40 (Uniprot: P25942-1), which can drive luciferase expression systems upon CD40 ligand/agonist antibody stimulation. In the absence of agonist antibody or CD40 ligand, the CD40 receptor is not activated, and luminescence signal is low. In the presence of agonist antibody or CD40 ligand, the CD40 pathway-activated luminescence can be detected in a dose-dependent manner. This reporter cell can also be used to test whether agonist antibodies enhance agonistic activity in an Fc纬R-dependent manner.

CD40 is a co-stimulatory receptor expressed on antigen-presenting cells (APCs) and B cells. It binds to CD40 Ligand (TNFSF5) on T cells, promoting APC activation, B cell proliferation, and antibody class switching. CD40 signaling is critical for germinal center formation and humoral immunity.
