The HEK293-Human HVEM (Luc)-Reporter Cell is engineered to express both the NF-kB signaling response element and the receptor full-length human HVEM (Uniprot: Q92956-1). When co-cultured with target cells expressing human BTLA, the BTLA/HVEM interaction drives NF-kB-mediated luminescence. Blocking the BTLA/HVEM interaction by either anti-BTLA or anti-HVEM antibodies results in a decrease in luminescence.

HVEM (Herpesvirus Entry Mediator, TNFRSF14) is a dual-function receptor expressed on immune cells. It binds to LIGHT (TNFSF14, activating) or BTLA/CD160 (inhibitory), modulating lymphocyte activation and immune tolerance. HVEM is involved in autoimmune diseases and tumor immune evasion.
