Introduction

This Jurkat reporter cell line expresses the NFAT response element driving luciferase expression and the full-length human PD-1 (Uniprot: Q15116) and LAG-3 (Uniprot: P18627-1) receptors. It can be used to evaluate the synergistic effect of anti-human PD-1 and anti-human LAG-3 antibodies. When co-cultured with target cells expressing human PD-L1 and MHCⅡ, the PD-1/PD-L1 and LAG-3/MHCⅡ interactions inhibit TCR signaling and NFAT-mediated luminescence. Blocking these interactions with anti-PD-1 or anti-PD-L1 and anti-LAG-3 antibodies releases the inhibitory signals, resulting in TCR activation and NFAT-mediated luminescence.

Application

Screen for anti-human PD-1 or/and anti-human LAG-3 antibody

Background

PD-1 and LAG-3 are both immune checkpoint receptors that inhibit T cell function, and their co-expression is associated with enhanced immune suppression in tumors.
