This Jurkat reporter cell line expresses the NFAT response element driving luciferase expression and the full-length human TIGIT receptor (Gene ID: 201633). It can be used to evaluate the potency of TIGIT blockade. When co-cultured with target cells expressing human CD155, the TIGIT/CD155 interaction inhibits TCR signaling and NFAT-mediated luminescence. Blocking the TIGIT/CD155 interaction with anti-TIGIT or anti-CD155 antibodies releases the inhibitory signal, resulting in TCR activation and NFAT-mediated luminescence.

TIGIT is an immune checkpoint receptor expressed on T cells and natural killer cells. It inhibits immune cell activation and plays a role in immune tolerance and tumor immune evasion.
