Hematopoietic progenitor kinase 1 (HPK1), also known as MAP4K1, is a serine/threonine protein kinase belonging to the MAP4K family. It plays a crucial role in immune signaling pathways by negatively regulating T cell receptor (TCR) and B cell receptor (BCR) signaling. HPK1 is primarily expressed in hematopoietic cells and immune cells, including T cells, B cells, macrophages, and dendritic cells. It acts as a negative feedback regulator of TCR signaling by phosphorylating SLP76 and Gads, leading to the downregulation of ERK signaling and the ubiquitination degradation of SLP-76, which results in decreased TCR signaling and T cell proliferation. HPK1 is considered a promising target for cancer immunotherapy, as inhibiting HPK1 can enhance T cell activity and improve anti-tumor immune responses.
